In another staged display of loyalty, Iago threatens Roderigo for telling Brabantio of Othello and Desdemona's marriage. Brabantio accuses Othello of bewitching his daughter into falling in love with him.
When Roderigo and Iago warn Brabantio that Desdemona has married Othello, he's upset and doesn't believe it's true. Roderigo and Iago tell Brabantio that his daughter is sleeping with Othello in order to upset Brabantio and further damage Othello out of jealousy and racism.
*Iago does the talking and tells Brabantio he will have black"animals" for grandchildren. *Roderigo confirms to Brabantio that his daughter did marry Othello. *Iago then tells Roderigo to go warn Othello that Brabantio is coming after him. Iago can't warn Othello because he works for Othello.
Iago, hidden by the night, proclaims that Brabantio's heart should be broken, as half of his soul is stolen. Iago declares to him that "an old black ram is tupping your white ewe."
As we know, Iago lists multiple (and incompatible) motives for seeking to destroy Othello (elsewhere, he says he hates Othello because he was passed up for a promotion), so it's entirely possible that Iago's the one who makes up the rumor about Othello and Emilia.
Othello is unaffected by Brabantio's potential threats regarding the marriage to his daughter because he currently needs to focus on the matters of war that the Duke of Venice has asked him to be a part of. According to Brabantio, why has Desdemona married Othello?

In 1.2, Brabantio is led to the Sagittary, where the newlyweds are found and there accuses Othello of using magic to bewitch his daughter. In 1.3, he brings Othello to trial before the Duke and once again accuses him of using witchcraft upon his daughter.
Iago here emphasises the biracial nature of the marriage, already showing his ability to manipulate people, in this case he is manipulating Brabantio, to believe in Iago's own opinions and in theory to eliminate all thoughts that Brabantio might of had of his own about the marriage.

Iago wants revenge on Othello because he is jealous of Cassio's promotion and jealous of Othello's suspected relationship with his wife. He then fuels Othello's own jealousy by leading him to believe that Desdemona and Cassio are in a relationship.
